The combinations of two words are typically used as part of a registration form to ensure the registrant is, indeed, human. Google is now replacing one of the words in some of its reCAPTCHA forms with photos gleaned from Street View service. Google says it uses these numbers internally to improve the accuracy of Street View and that pulling them into reCAPTCHAs is part of an "experiment" to "determine if using imagery might also be an effective way to further refine our tools for fighting machine and bot-related abuse online."

In other words, Google's bots are already capable of decoding these numbers, which makes this all sound like a bit of a challenge to the rest of the OCR-loving coders in the world. Any takers?

Such work requires active attention to what we are doing, why we are doing it and what is happening as a result.? It requires a mind to gain understanding and make good decisions, and the will to lock onto worthwhile goals.? And this feels like hard work.

Which is why, in my experience, we need two other ?goods? to help us do good work.? We need good friends and good questions.? Good friends are not just acquaintances or buddies with whom we have a lot in common and whose company we enjoy.? Good friends are often quite different from us, and those differences can make them think or act in unexpected ways.? They can sometimes seem like strangers.? Good friends offer us a different viewpoint and, sometimes, a critical perspective on our action.

We don?t just fall into these kinds of relationships.? Friendship like this cannot be taken for granted.? I found good friends in my PhD supervision group.? They challenged my assumptions and wouldn?t let go when they felt I needed to dig deeper in my reflections.? Good friends have the courage to talk back, to push against my hubris.? Good friends, in words from the Book of Hebrews, provoke one another to good work.

The second ?good? intrinsic to good work is good questions.? The point of a good question is not a correct answer.? In this sense the question is just a means to an end.? But good questions don?t have simple answers.? We can, of course, treat interruptions to our action in a simple way by asking ?what can I do differently??? There may be other ways of doing things and we can try them.? But this may not be a ?good? question.? A good question will probe deeper.? Good questions push us to pay closer attention.? They tease out our assumptions about the ends and not just the means.? They suggest different horizons.? They can transform black and white into colour, and help us hear the harmonies behind a simple melody.? Good questions transform looking into real seeing, and hearing into true listening.

Good work doesn?t just happen.? It calls for informed, courageous choices.? I make no claim to do good work but I aspire to it and I know the importance of good friends and good questions in achieving it.? As a learning community, we hope that GoodWork will provides a space for us to explore this, each of us in different ways, in our daily practice.

And so, some questions for us to explore:

Patients with irritable bowel syndrome (IBS) have a significantly greater prevalence of early adverse life events , including general trauma as well as physical, emotional and sexual abuse, according to a new study in Clinical Gastroenterology and Hepatology, the official journal of the American Gastroenterological Association.

"Various types of early adverse events are associated with the development of irritable bowel syndrome, particularly among women," said Lin Chang, MD, of the David Geffen School of Medicine at the University of California Los Angeles and lead author of this study. "Addressing early adverse events and associated psychological symptoms in these patients is important and may help guide management approaches that reduce symptoms and improve overall well-being."

Although associations between an abuse history and IBS have been reported before, in the current study, researchers aimed to assess simultaneously the association of a range of traumatic events ? not limited to abuse ? with IBS and the impact of differences in gender and psychological factors on these associations. Men and women with IBS who were 18 years of age and older were recruited primarily from community advertisements in the greater Los Angeles area. Early adverse life events were evaluated in 294 IBS patients (79 percent were women) and 435 controls (77 percent were women). Validated questionnaires assessed gastrointestinal, psychological and somatic symptoms.

Compared with controls, IBS patients reported a higher prevalence of general trauma, physical punishment, emotional abuse and sexual events. These events included witnessing violence, mental illness in the family, emotional abuse, and being forced to touch intimate parts of a person's body or have genital sex. These significant differences were observed mainly in women. Emotional abuse was the strongest predictor of IBS. Further, the strength of the relationship between the early trauma and IBS was reduced after controlling for the presence of psychological and other non-gastrointestinal symptoms.

Early adverse life events refer to traumatic experiences during childhood encompassing physical, sexual or emotional abuse, as well as discordant relationships with a primary caretaker, or the loss of a parent. These events appear to be associated with an increased vulnerability toward developing functional gastrointestinal disorders, including IBS.

US jobless claims fall to lowest level in 4 years

WASHINGTON (AP) ? The number of people seeking U.S. unemployment benefits dropped last week to the lowest level in four years, adding to evidence that the job market is strengthening.

Applications for weekly unemployment benefits fell by 5,000 to a seasonally adjusted 359,000, the Labor Department said Thursday. That's the fewest applicants since April 2008. The four-week average, a less volatile measure, declined to 365,000 ? the fewest for that measure since May 2008.

When unemployment benefit applications drop consistently below 375,000, it usually signals that hiring is strong enough to lower the unemployment rate. The decline has coincided with the best three months of hiring in two years.

The department made annual revisions to the past five years of data, which increased the number of applications in recent months and showed a slower decline. Still, even after the revisions, applications have fallen roughly 12 percent over the past six months.

Most economists still expect another strong month of hiring in March.

"The trend remains unambiguously downwards," said Ian Shepherdson, an economist at High Frequency Economics. "We think the rate of decline ... is slowing ... but they are still consistent with robust, sustained payroll gains."

Separately, the U.S. economy expanded at a solid pace in the final three months of last year, but growth is expected to slow in the current quarter.

The economy grew at annual rate of 3 percent in the fourth quarter, the Commerce Department said. But economists forecast it likely dipped below 2 percent in the current quarter.

Businesses have been restocking their shelves at a slower pace and shipping fewer long-lasting manufactured goods. In addition, Europe's debt crisis and slower growth in Asia have reduced demand for U.S. exports.

Stronger hiring in the first two months of the year probably hasn't offset those weaknesses. That's because Americans' pay has barely kept pace with inflation while gas prices have spiked. So consumer spending, which accounts for 70 percent of economic activity, probably hasn't increased much from the end of last year.

Most economists expect growth to pick up later this year, as more hiring lifts the economy.

From December through February, employers added an average of 245,000 jobs per month. That has pushed down the unemployment rate to 8.3 percent, the lowest in three years.

Companies are hiring more workers because the economy is picking up. The economy grew at an annual rate of 3 percent in the final three months of last year. That was better than the 1.8 percent rate in the previous quarter.

The number of people receiving unemployment benefits also fell. More than 7.1 million laid-off workers received unemployment aid in the week ended March 10, the latest data available. That's about 130,000 lower than the previous week.

There are other signs the economy is steadily recovering. Consumers remained confident in March, even as gas prices jumped.

And the battered housing market is showing small signs of improvement. January and February comprised the best winter for sales of previously occupied homes in five years, according to the National Association of Realtors.

One concern is that rising gas prices will force consumers to cut back on discretionary spending. That could weigh on economic growth and slow hiring. The Federal Reserve says it expects oil and gas prices to temporarily boost inflation but predicts that longer-term inflation should remain stable.

The job market still has a ways to go to fully recover from the Great Recession. More than 12.8 million people remain unemployed and the economy still has 5 million fewer jobs than before the downturn.

But the more robust job market has caused some so-called "discouraged workers" to start looking again. The work force rose by nearly a half-million in February.

A Conversation With Macy Gray

Mike Ragogna: Hello to Macy Gray.

Macy Gray: Hi!

MR: Macy, first of all, how have you been, everything's great?

MG: Yeah, everything is really good.

MR: Great. This time out, you decided to do an album of covers called, well, Covered. What were you thinking with this one?

MG: I've always wanted to do a covers album, we finally did one, and it was a big deal about what kind of songs to do. I thought there were a couple things that would be obvious like if I did old soul records or R&B covers, so we did rock records because I'm a big rock 'n' roll (fan), and then it was a matter of what rock records to do. We mostly did modern stuff that just came out last year and goes all the way back to the Eurythmics, a 1985 song. We did "Nothing Else Matters" by Metallica, so it's like the past, say, 25 years of really good rock 'n' roll songs that we covered.

MR: Yeah, you covered Metallica, you did "Teenagers" by My Chemical Romance, and also you did "Creep" by Radiohead.

MG: We've been doing "Creep" live for a long time, so it made sense to do that one.

MR: Yeah, there's also a very cute "song" on here, "Lalala," by Teaching the Kids. What's that track's approach about?

MG: We actually have these really hilarious skits on the album. The kids on one of them are friends of my daughter, friends from high school. We have Nicole Scherzinger on a skit, she's doing these crazy imitations, the funniest things you'll ever hear. Then we have MC Lyte and Jay-Z upping the enthusiasm. It's a very entertaining album, but the music is awesome. We did everything live, everybody plugged in and recorded at the same time. It's live like we did it back in the old days, produced by Hal Willner and Sue, and it's just a great album. I love listening to it.

MR: You also covered "Wake Up" by Arcade Fire, the Eurythmics, "Here Comes the Rain Again," and a cover of Sublime's "Smoke Two Joints." How are you relating to these covers? Are there memories associated?

MG: Yeah, definitely, there are always memories attached to the songs. When you hear a song, I always remember a moment that you have with it. A lot of it mostly was lyrics that were very important to me, and a lot of it was songs that secretly I would like to pull off. There are a lot of great songs out there, but some of them should just be left alone, and some of them, I didn't sound very good on at all. So, those were the ones that came out the best and that I felt I could interpret and make my own.

MR: Macy, you have a history of being self-deprecating about your voice, when everybody loves it.

MG: It's hilarious, I know.

MR: (laughs) Let's get a little history lesson as far as Macy Gray. What got you into music?

MG: I studied piano lessons when I was 7, I did that until I was 14, but I hated it so much. When I got to college, I met a boy, he was a guitar player. He had a studio in his room, and I used to do music all day just so I could be around him. I got really obsessed with music all of a sudden - I started writing, I sang in his band, and one thing led to another, and I started my own band a couple years later. We were playing at The Roxy in Los Angeles and an A&R guy from Atlantic signed me there. That record didn't come out, but I did a record on Sony two years later called How Life Is and that did really well.

MR: Right, and you of course wrote the Black Eyed Peas song "Love Won't Wait."

MG: Yes.

MR: Then there was "I Try." Were you surprised it was such big hit?

MG: I was really shocked. We were out on the road and we had gone to Europe. I was in Europe for the first time, I had three kids with me. All my buddies were in my van, and the label bought me a new car, so I was in heaven. I didn't really need anything else. My manager calls me and says that the song was #1. I was really excited but I felt busy on the road, it was my first #1, but I really didn't know what it meant. I didn't really go crazy until I got home and got mauled in New York, and that's when I really realized that people knew who I was. But I wasn't really shocked when it hit, though.

MR: And you also must have been in heaven when you get your Grammy nomination.

MG: Yeah, all that, it was like one shock after another. I think the cool thing was that I wasn't expecting anything. My first week of sales, I sold 9,000 copies and I was ecstatic because I had never sold a record before. Everything that happened after was just juice. I wasn't really expecting any of it.

MR: The other part of this history lesson is that you've been in a few films, and one of them was Spider-Man where you played yourself.

MG: That was difficult, I wasn't really prepared for that role.

MR: (laughs) And you were also in Training Day.

MG: Yes, Training Day. We did Idlewild, I did Domino with Tony Scott and Keira Knightley. I've done some really good movies - Lackawanna Blues... I've been lucky.

MR: Yeah, you've been blessed. Getting back to Covered, I'm imaging over the years, you were learning a lot of the songs at the time that they were hits, right?

MG: Right.

MR: So when you sat back and listened to Covered top to bottom, was it kind of like looking at a photo album?

MG: Yeah, I hadn't thought of it that way before, but I definitely have a lot of visuals that come to mind when I hear "Hear Comes the Rain," or definitely the first time I listened to "Nothing Else Matters." I remember that. All of them. "Bubbly" is on there by Colbie Caillat and I really love that song. I think that's a personal thing with everybody. When you have a song, it brings something to mind.

MR: How do you approach your arrangements, are you the main person coming in there with ideas?

MG: No, I have a really intense band and they do most of the arrangements, even though I direct a little bit. I say "I like this, I don't like this," but they really come up with the best ideas, way better than mine.

MR: Are you entrenched in each song to the point where you are the character of the song, maybe losing yourself in it?

MG: Oh yeah, always. I always seem to take them to heart. The cool thing about those songs is that I had a personal connection to them, so it wasn't hard. I do think it's important that when you sing a song that it means everything to you, because if it doesn't, it's definitely going to affect the way you sing.

MR: What's your favorite song on Covered?

MG: I really like "Love Lockdown," the mash-up. I love that song.

MR: Yeah, the Nina Simone song. Is she one of your favorite artists?

MG: Yeah, absolutely, I learned a lot from her. She has this cover of Frank Sinatra's "My Way"; it's big-band jazz, it's crazy. Actually, when I heard that, that gave me the confidence to do my record because she made it completely her own and I think a lot of people do covers and think that they have to be loyal to the song and do things over. But after I saw that, I thought, "I'm going to go off and to do my own thing." That's really what made the record happen.

MR: So that was an inspiration on how you would approach covers. Let's take the other side of that question, about your own original material. What is your creative process?

MG: I just lock-up myself in the studio and just start writing. You just pull your heart out, really, if you're in a tangle. That's the best thing you can do.

MR: Do you find yourself working on piano, or is it in your head and you have to get to an instrument to write it down?

MG: Yeah, a lot of songs pop into my head all the time. If I don't remember them, I figure that maybe they weren't that great. The ones I remember, those are the ones I usually record. I play a little bit of guitar, I know about six chords on the guitar, so anytime I write a song with music, I use the guitar because it's portable. But I wrote "I Try" on the guitar. At the time, those were the only three chords I knew, A and E.

MR: Macy, do you have other favorites of your originals?

MG: My favorite songs that I've ever done are probably "Caligula" and "I've Committed Murder," both from the first album. I have a song on my last album called "Sellout," and a song called "Let You In," I love that.

MR: What advice might you have for new artists?

MG: I'm not a good advice-giver, actually. But I would just tell anybody if you really want to do it, you just have to pull your heart out at all times and be completely open. Scream when you want to, and write when you want to, and you should be alright.

MR: Is that what you would tell the young Macy Gray?

MG: Yes. I would say, "Do and say the things that you truly feel."

MR: That's basically what you've been doing your whole career, right?

MG: Yeah there's no other way, really, for me anyway.

MR: OK, so also as far as a new project beyond Covered, have you been collecting your original material for a new album?

MG: Yeah, that's ready to go already because I write all the time so I have tons of songs ready to go. I actually wrote a song last night, which I wish I could put on the radio right now.

MR: Yeah, we wouldn't want you to hum a little of it or anything. (laughs)

MG: I can't hum. (laughs) But it's a great song, I can't wait for it to come out.

MR: This is beautiful. Macy Gray, do you have any words of wisdom for us?

MG: Words of wisdom?

MR: What about anything in the news, is anything catching your eye?

MG: I haven't watched the news in the last few days, but you know what happened is that there was a situation that happened in the news a little while ago. So much information was wrong that I really got frustrated with the news and haven't watched since.

MR: What happened?

MG: It was the whole Whitney Houston thing. I just saw a lot of the information that was going around, and so much of it was wrong, from sources that we consider really on target. After I saw that, I've just been turned off. I'm a big news-watcher. I love the news and politics, but after that, I've just been disenchanted with that whole thing and haven't watched it for a while.

MR: Yeah, I guess it's exaggeration in order to get you excited so you stay tuned to the news.

MG: Yeah, it's crazy. I read quite a bit, but I just haven't watched the news in a while.

MR: Macy, thank you for all your time, I'd love to do this again in the future, maybe with your next new album.

MG: Anytime.

A Conversation with 'ZinePak Founders Kim Kaupe and Brittany Hodak

Mike Ragogna: How did 'ZinePak come about, the idea, how did it start, what inspired you??

Brittany Hodak: I started my career in the music industry, and Kim started hers in publishing. We were both incredibly passionate about the idea of combining print and music to create an engaging, multi-sensory experience that digital downloads and tradition CDs can't deliver. 'ZinePak is a little counter to the "digital" only online blog magazine trend.

MR: What made you decide to do something more traditional with music and a magazine??

Kim Kaupe: Fans still crave an experience with their favorite artist - something they can see, touch, and engage with. Creating a piece that includes limited-edition merch items as well as photos, edit and music was an answer to the incessant demand from fans to experience more. We also feel that these pieces can be put on your coffee table, shown to friends and passed around unlike a release on a Kindle or an iPad.??

MR: What was the first 'ZinePak and was it a challenge to put it together??

BH: Our first 'ZinePak was for the release of Kidz Bop 19 in January of 2011. We've done two more projects with the Kidz Bop brand since.
??
MR: How did the next 'ZinePak come about??

KK: Our second project, the 2011 ACM Awards 'ZinePak, was brought to us by our partners at Walmart. They really championed our idea of connecting consumers with an Awards property, and they introduced us to the great team at the Academy. ??

MR: Have the artists been easy to work with?

KK: All of the artists that we have worked with have been really excited which has been a fantastic experience. Fans can tell when a manager or record label releases a statement - they are much smarter than anyone gives them credit for. When artists realize that this is an opportunity to communicate with their fans in a long-format multi-sensory way, the ideas for content and items start pouring out of them.?

MR: Is it easier to do a 'ZinePak based on one artist or is there something fun for you about putting together a compilation 'ZinePak??

BH: It depends. The great thing about 'ZinePaks is that every one is totally different, and the content inside is tailored to the release and to the fans. They're all a blast to put together, and each project presents its own challenges and opportunities.?

MR: Where can you buy 'ZinePak?

BH: The ACM 'ZinePak is available exclusively at Walmart.

MR: Why Walmart?

BH: Walmart is a great partner because they're the number one seller of physical music product in America, and they have a huge market share for country releases. We've also done 'ZinePaks for artists' tours and non-traditional retail partners. ??

MR: What's next for the brand?

KK: On April 3rd, a 'ZinePak version of Rascal Flatt's new album Changed will be released. We also just confirmed this morning - and can announce exclusively here - that we will be creating a 'ZinePak for The Beach Boys. They are a personal favorite, so I am over the moon! That will be in stores May 1, just in time to help commemorate the band's 50th Anniversary.

MR: What is your dream 'ZinePak to do, like is there an artist or maybe an award show you would like to do??

BH: Hands down...Garth Brooks. I have so much respect for him as an artist and a performer, and working with him is my dream.

KK: The Super Bowl. Or maybe an NCAA championship game.??

MR: What are you brand goals??

KK: It's a simple sentence and a big goal: Redefine the way fans experience and engage with recorded entertainment. ??

MR: Is there a digital 'ZinePak headed our way soon or no?

BH: 'ZinePaks are all about creating an engaging experience for fans. We've discussed digital releases and may put something out in the future, but for now we love providing the experience of taking the shrink-wrap off of a product, opening it and finding fun physical elements inside - like magnets, trading cards, stickers, etc.???

MR: By the way, was the ACM 'ZinePak a challenge to make in any way?

BH: The challenge we faced with the ACM 'ZinePak was getting it to market quickly. We went to print about 10 days after the nominees were announced, so that didn't leave much time to do interviews and gather content. We were lucky to have amazing partners at the Academy and record labels who helped us get everything together quickly.

MR: Well, Award shows seem more conducive to compilations.

KK: I think award shows are great for compilations because fans already know that their favorite artists are going to be performing in one place. MR: Do you feel Country music fans tend to buy more traditional/physical product??

BH: Yes. Compared to other genres, physical CDs account for a larger percentage of total sales. That's in part, I think, because country fans like to feel connected to the artists they listen to, which is why 'ZinePak is a perfect fit for the audience.??

MR: Last question, what else would you like people to know about 'ZinePak and perhaps this release??

KK: We are a company built on the principle that you have to do what you love. We love making 'ZinePaks, and we love creating meaningful experiences for artists to share with their fans. We hope fans can see all the love and care that we (and the artists!) put into this release for them.

MR: What advice do you have for new artists??

KK: Grow a very thick skin and get ready for anything that comes your way. Sometimes it is not the "best" artists musically that hit it big, but the ones who stick it out the longest and know how to keep getting up after being knocked down.??

The rise of the virtual business
I?ve been involved recently with advising a client on setting up a new business. It was an interesting exercise in the art of the possible ? how to build a virtual business with almost no infrastructure or fixed costs and limitless profit potential.

Background

At the outset, the client thought in terms of renting offices, buying stock, employing staff and having to raise money to turn the vision into a reality.

As we progressed, that was proved not to be the case and the idea was brought to market much more rapidly and at much lower cost and risk than the client originally envisioned.

Potent forces

To drive the client?s idea forwards, we combined three potent forces. They were:

  • Relentless focus on the customer
  • Use of contractors and consultants to get things done fast
  • Outsourcing everything possible

Customer focus

The client?s vision was strong. A big part of the challenge was to keep the client?s focus on customer value without the distractions of the traditional business model.

Building an almost purely virtual business took all the day-by-day implementation issues away and enabled the client to focus on how best to create and market their product.

The methods involved also had the interesting side effect of providing a lot of the capital required to start the business.

Use of contractors and consultants

Rather than employ staff full time, the client engaged a number of contractors and consultants to get design work completed, a go-to-market plan designed and implemented and set up the basic business processes required.

Each contractor or consultant was employed on day rates to deliver defined deliverables in a defined timescale. This resulted in no ongoing employment liabilities or costs ? critical for a start up business.

Outsourcing everything

With the business basics established, everything was then outsourced. This included business processes, manufacturing and distribution. Everything was delivered against contracts specifying pricing and performance based on volume.

This had the effect of moving a big chunk of the risk in a start-up company to outside specialist providers, and also effectively provided working capital to get started.

The result?

It worked. The client?s vision was brought to market rapidly with minimal investment.

The consequences for traditional businesses are frightening. Their very size and scale makes them inflexible and unable to react in the timescales they are being forced into.

Novel compound halts tumor spread, improves brain cancer treatment in animal studies

ScienceDaily (Mar. 28, 2012) ? Treating invasive brain tumors with a combination of chemotherapy and radiation has improved clinical outcomes, but few patients survive longer than two years after diagnosis. The effectiveness of the treatment is limited by the tumor's aggressive invasion of healthy brain tissue, which restricts chemotherapy access to the cancer cells and complicates surgical removal of the tumor.

To address this challenge, researchers from the Georgia Institute of Technology and Emory University have designed a new treatment approach that appears to halt the spread of cancer cells into normal brain tissue in animal models. The researchers treated animals possessing an invasive tumor with a vesicle carrying a molecule called imipramine blue, followed by conventional doxorubicin chemotherapy. The tumors ceased their invasion of healthy tissue and the animals survived longer than animals treated with chemotherapy alone.

"Our results show that imipramine blue stops tumor invasion into healthy tissue and enhances the efficacy of chemotherapy, which suggests that chemotherapy may be more effective when the target is stationary," said Ravi Bellamkonda, a professor in the Wallace H. Coulter Department of Biomedical Engineering at Georgia Tech and Emory University. "These results reveal a new strategy for treating brain cancer that could improve clinical outcomes."

The results of this work were published on March 28, 2012 in the journal Science Translational Medicine. The research was supported primarily by the Ian's Friends Foundation and partially by the Georgia Cancer Coalition, the Wallace H. Coulter Foundation and a National Science Foundation graduate research fellowship.

In addition to Bellamkonda, collaborators on the project include Jack Arbiser, a professor in the Emory University Department of Dermatology; Daniel Brat, a professor in the Emory University Department of Pathology and Laboratory Medicine; and the paper's lead author, Jennifer Munson, a former Fulbright Scholar who was a bioengineering graduate student in the Georgia Tech School of Chemical & Biomolecular Engineering when the research was conducted.

Arbiser designed the novel imipramine blue compound, which is an organic triphenylmethane dye. After in vitro experiments showed that imipramine blue effectively inhibited movement of several cancer cell lines, the researchers tested the compound in an animal model of aggressive cancer that exhibited attributes similar to a human brain tumor called glioblastoma.

"There were many reasons why we chose to use the RT2 astrocytoma rat model for these experiments," said Brat. "The tumor exhibited properties of aggressive growth, invasiveness, angiogenesis and necrosis that are similar to human glioblastoma; the model utilized an intact immune system, which is seen in the human disease; and the model enabled increased visualization by MRI because it was a rat model, rather than a mouse."

Because imipramine blue is hydrophobic and doxorubicin is cytotoxic, the researchers encapsulated each compound in an artificially-prepared vesicle called a liposome so that the drugs would reach the brain. The liposomal drug delivery vehicle also ensured that the drugs would not be released into tissue until they passed through leaky blood vessel walls, which are only present where a tumor is growing.

Animals received one of the following four treatments: liposomes filled with saline, liposomes filled with imipramine blue, liposomes filled with doxorubicin chemotherapy, or liposomes filled with imipramine blue followed by liposomes filled with doxorubicin chemotherapy.

All of the animals that received the sequential treatment of imipramine blue followed by doxorubicin chemotherapy survived for 200 days -- more than 6 months -- with no observable tumor mass. Of the animals treated with doxorubicin chemotherapy alone, 33 percent were alive after 200 days with a median survival time of 44 days. Animals that received capsules filled with saline or imipramine blue -- but no chemotherapy -- did not survive more than 19 days.

"Our results show that the increased effectiveness of the chemotherapy treatment is not because of a synergistic toxicity between imipramine blue and doxorubicin. Imipramine blue is not making the doxorubicin more toxic, it's simply stopping the movement of the cancer cells and containing the cancer so that the chemotherapy can do a better job," explained Bellamkonda, who is also the Carol Ann and David D. Flanagan Chair in Biomedical Engineering and a Georgia Cancer Coalition Distinguished Cancer Scholar.

MRI results showed a reduction and compaction of the tumor in animals treated with imipramine blue followed by doxorubicin chemotherapy, while animals treated with chemotherapy alone presented with abnormal tissue and glioma cells. MRI also indicated that the blood-brain barrier breach often seen during tumor growth was present in the animals treated with chemotherapy alone, but not the group treated with chemotherapy and imipramine blue.

According to the researchers, imipramine blue appears to improve the outcome of brain cancer treatment by altering the regulation of actin, a protein found in all eukaryotic cells. Actin mediates a variety of essential biological functions, including the production of reactive oxygen species. Most cancer cells exhibit overproduction of reactive oxygen species, which are thought to stimulate cancer cells to invade healthy tissue. The dye's reorganization of the actin cytoskeleton is thought to inhibit production of enzymes that produce reactive oxygen species.

"I formulated the imipramine blue compound as a triphenylmethane dye because I knew that another triphenylmethane dye, gentian violet, exhibited anti-cancer properties, and I decided to use imipramine -- a drug used to treat depression -- as the starting material because I knew it could get into the brain," said Arbiser.

For future studies, the researchers are planning to test imipramine blue's effect on animal models with invasive brain tumors, metastatic tumors, and other types of cancer such as prostate and breast.

"While we need to conduct future studies to determine if the effect of imipramine blue is the same for different types of cancer diagnosed at different stages, this initial study shows the possibility that imipramine blue may be useful as soon as any tumor is diagnosed, before anti-cancer treatment begins, to create a more treatable tumor and enhance clinical outcome," noted Bellamkonda.
